How to deploy new Distribution Point in a second site? Does it require secondary Configuration Manager within the second site?

When you say second site, you mean another office location ?. If yes, you just need to setup a server, install the DP role on that server.

So this does not require a secondary sccm site but only configuring the new server with the DP Role?

Yes, Secondary sites are useful when you have remote site and you want to transfer content and client data across low-bandwidth networks. If you have got sufficient bandwidth, you can always have a distribution point for every site.
